Mapping vesicle shapes into the phase diagram: A comparison of experiment and theory. Abstract Phase-contrast microscopy is used to monitor the shapes of micron-scale fluid-phase phospholipid-bilayer vesicles in aqueous solution. At fixed temperature , each vesicle undergoes thermal shape fluctuations. We are able experimentally to characterize the thermal shape ensemble by digitizing the vesicle outline in real time and storing the time-sequence of images. Analysis of this ensemble using the area-difference-elasticity (ADE) model of vesicle shapes allows us to associate (map) each time-sequence to a point in the zero-temperature (shape) phase diagram. Changing the laboratory temperature modifies the control parameters (area, volume, etc.) of each vesicle, so it sweeps out a trajectory across the theoretical phase diagram. It is a nontriv-ial test of the ADE model to check that these trajectories remain confined to regions of the phase diagram where the corresponding shapes are locally stable. In particular, we study the thermal trajectories of three prolate vesicles which, upon heating, experienced a mechanical instability leading to budding. We verify that the position of the observed instability and the geometry of the budded shape are in reasonable accord with the theoretical predictions. The inability of previous experiments to detect the " hidden " control parameters (relaxed area difference and spontaneous curvature) make this the first direct quantitative confrontation between vesicle-shape theory and experiment.

Phase diagrams and shape transformations of toroidal vesicles

Shapes of vesicles with toroidal topology are studied in the context of curvature models for the membrane. For two simplified curvature models, the spontaneous-curvature (SC) model and the bilayer-couple (BC) model, the structure of energy diagrams, sheets of stationary shapes and phase diagrams are obtained by solving shape equations for axisymmetric shapes. Three different sheets of axisymmet...

Mapping Activity Diagram to Petri Net: Application of Markov Theory for Analyzing Non-Functional Parameters

The quality of an architectural design of a software system has a great influence on achieving non-functional requirements of a system. A regular software development project is often influenced by non-functional factors such as the customers' expectations about the performance and reliability of the software as well as the reduction of underlying risks. The evaluation of non-functional paramet...

Gel phase vesicles buckle into specific shapes.

Osmotic deflation of giant vesicles in the rippled gel phase P(β') gives rise to a large variety of novel faceted shapes. These shapes are also found from a numerical approach by using an elastic surface model. A shape diagram is proposed based on the model that accounts for the vesicle size and ratios of three mechanical constants: in-plane shear elasticity and compressibility (usually neglect...

Gravity-Induced Shape Transformations of Vesicles

We theoretically study the behavior of vesicles filled with a liquid of higher density than the surrounding medium, a technique frequently used in experiments. In the presence of gravity, these vesicles sink to the bottom of the container, and eventually adhere even on non attractive substrates. The strong size-dependence of the gravitational energy makes large parts of the phase diagram access...

Pressure-temperature phase diagram for shapes of vesicles: A coarse-grained molecular dynamics study

Coarse-grained molecular dynamics simulations are performed to obtain the phase diagram for shapes of a vesicle with a variation in temperature and pressure difference across the membrane. Various interesting vesicle shapes are found, in particular, a series of shape transformations are observed for a vesicle with an initial spherical shape, which changes to a prolate shape, then an oblate shap...
